Does Putting Niacinamide on Your Skin Raise NAD⁺?

October 14, 2026 2026년 10월 14일 14 octobre 2026

Quick answer: Not in any simple or predictable way. Niacinamide (nicotinamide, NAM) is a building block the cell can use to make NAD⁺, but the first step of that conversion is controlled by an enzyme that sets the pace, and NAD⁺ is being spent as fast as it is made. So the amount of niacinamide you put on your skin and the amount of NAD⁺ inside your skin cells are two different numbers. Niacinamide is a well-studied cosmetic ingredient in its own right; it does not need an NAD⁺ story to be worth using, and an NAD⁺ story is not a reason to use more of it.

This is part one of a four-part series on niacinamide, NAD⁺ and DIY skincare.

The claim

A version of this circulates widely: niacinamide turns into NAD⁺, NAD⁺ is the molecule behind cellular energy and skin ageing, so a cheap niacinamide powder does the same job as an expensive NAD⁺-focused product. Each link in that chain needs checking on its own.

The conversion is not automatic

In the cell's recycling route, called the salvage pathway, the sequence is:

Niacinamide (NAM) → NMN → NAD⁺

The first step is carried out by the enzyme NAMPT (nicotinamide phosphoribosyltransferase), which attaches niacinamide to a sugar-phosphate donor called PRPP to make NMN (Wang 2006). In mammals, NAMPT is the rate-limiting enzyme of this route (Revollo 2007). Rate-limiting means the enzyme, not the supply of niacinamide, sets how fast NMN can be made. Once the enzyme is working at capacity, adding more niacinamide does not push more through; it also depends on the cell having enough PRPP and energy available for the reaction.

NAD⁺ is spent as fast as it is made

NAD⁺ is not stored like a reserve. It carries electrons in energy metabolism, and it is also consumed outright by enzymes such as sirtuins, PARPs (which use it during DNA repair) and CD38 (Covarrubias 2021). The level in a cell at any moment is the balance between making and spending. A change in supply is only one side of that balance, and the spending side rises with age, inflammation and UV stress.

What the skin-cell studies actually show

The direct evidence in skin cells is small and does not point one way.

  • In cultured human keratinocytes, nicotinamide and its derivatives did not raise NAD⁺ levels, while nicotinic acid (a different form of vitamin B3, using a different enzyme route) raised them about 1.3-fold (Oyama 2024).
  • In another keratinocyte study, niacinamide did increase NAD⁺, alongside markers of keratinocyte differentiation (Nisbet 2019).

Both are cells in a dish, at concentrations chosen by the researchers, without a stratum corneum in the way. Neither measured NAD⁺ in living human skin after a cosmetic was applied. The fair summary is that whether topical niacinamide raises NAD⁺ in the skin of a person is not established.

Absorption, NAD⁺ and visible change are separate steps

Niacinamide does cross the stratum corneum, and how much crosses depends on the formula: in human skin it permeated about twice as well at pH 7.4 as at pH 5.0 (Sjöberg 2026). But each of the following is a separate question with its own evidence:

  1. Does the ingredient get through the outer layer?
  2. Is it taken up and converted inside living cells?
  3. Does the cell's NAD⁺ level actually rise, given what is being consumed?
  4. Does that change produce something you can see or feel on the skin?

A yes to the first is not a yes to the last.

What niacinamide is good for, without the NAD⁺ story

Niacinamide earns its place on its own record. In a 12-week, double-blind, split-face study of 50 women, 5% niacinamide improved the look of fine lines, uneven spots, red blotchiness and sallowness compared with its vehicle (Bissett 2005). It is also associated with lower water loss through the skin and better hydration of the outer layer (Gehring 2004). Those results come from studying the ingredient on skin, not from assuming what happens to NAD⁺. For tone specifically, see can niacinamide help uneven skin tone.

The rest of this series

  • Part 2: Is DIY niacinamide powder mixed with water safe?
  • Part 3: Why you shouldn't mix niacinamide powder into your serum or moisturizer
  • Part 4: Is topical NMN better than niacinamide for NAD⁺?

Where the evidence runs out

The enzyme biology (Wang 2006; Revollo 2007; Covarrubias 2021) is well established but comes mostly from non-skin tissues and models. The skin-specific NAD⁺ data are two cell-culture studies with opposite results (Nisbet 2019; Oyama 2024). The clinical niacinamide studies measured appearance, not NAD⁺, and were run by a single manufacturer's research group. This article is educational; it is not a diagnosis and not a substitute for care from a physician or dermatologist.

(NAM) → NMN → NAD⁺

Niacinamide (NAM) → NMN → NAD⁺
